How to digest tetrads with Zymolyase

1. Pick a sporulated culture with a toothpick
2. Add spores to 50 µl dH2O
3. Add 1ul of 5 U/µl Zymolyase (E1004B; Zymo Research) into the culture
4. Leave at room temperature for 5 minutes. Check the efficiency of Zymolyase under the microscope.
5. Repeat step 4 until the asci are digested to the correct level.
6. Add 150 µl dH2O
7. Spread a small amount of digested cells on plates
