Size
As well as being big enough to accept the ROTOR+ the installation should also be considered. The loading port is usually at one end only. Make sure that the opening is big enough to accept the ROTOR+.

 

Installation
The ROTOR+ weighs approximately 110Kg and is back heavy. If the opening is only at one end it may be quite challenging to get it safely through the loading port and placed in the correct position.

 

Glove ports
Situated at the back and front of the chamber are helpful for so when loading it may be necessary to move the bench it is sat on away from any walls.

 

 

 

 

PIXL is the most popular product that customers fit into an anaerobic chamber.

Considerations Size – As well as being big enough to accept the PIXL the installation should also be considered. The loading port is usually at one end only. Make sure that the opening is big enough to accept the PIXL.

 

PIXL Size Width: 640mm Depth: 720mm Height: 800mm Installation – The PIXL will probably need to be tilted to get it through the door and into the tent. This makes installation tricky but not impossible.

 

Location in the chamber – Unless using a light curtain the PIXL needs to have space in front to open the door far enough to load plates. The door width is approximately 60cm. This may restrict how far into the PIXL the customer can reach to load plates.

 

A light curtain – Advised for use in an anaerobic chamber or automated workflow (this needs to be specified when ordering the PIXL).

 

Retrofit of a light curtain is not possible on-site. We do not recommend the removal of the door as this will affect the warranty and introduce Health and Safety issues. Singer Instruments will not be able to assist in modifying or using the PIXL with the door removed.

 

Dump drawer
Needs to be able to be removed for emptying and cleaning so again the PIXL has to have a gap at the front.

 

The PickUpLine cabinet
At the back right of the PIXL. This will be an extreme reach from the front. A glove port in the right location at the back of the chamber may help but it will still be a difficult operation to change it.

 

The operating area
Where the plates are loaded is quite high up on the PIXL so the glove ports should be placed in a suitable location and the bench height may need to be adjusted. Computer location – the computer for the PIXL is fitted inside the PIXL itself but the monitor, keyboard, and mouse should be located outside of the chamber.

 

Cables
Allow for cables to exit the PIXL. The computer and mouse are wireless so will not need external cables. Extension cables may be required. The main ones are: Power HDMI – to the external monitor Ethernet cable for remote access and/or for local network storage

 

 


The PhenoBooth+ will work within an anaerobic chamber without any major problems

Power cable
A powered extension inside the chamber may be required with a sealed outlet from the chamber. If a direct connection is required to an external power socket a longer power lead may be required.

 

Computer location
We recommend placing the computer, keyboard, and mouse outside of the chamber.
The cable to the computer will also need a sealed outlet from the chamber.

 

Location
A small gap at the front of the PhenoBooth+ for loading/ unloading plates and filters
Consider how plates will be loaded and removed from the chamber.
An extra 1 or two ports nearer the airlock may be useful.

Glove ports

One in line with the Dump Drawer to dispose of used PickUpLine.

2 or 3 ports level with the top of the operating area of the PIXL – to allow access to loading the plates. As mentioned previously with the door in place access to the inside of the PIXL is limited and reaching in far enough to place the plates in the rear target plate bays may not be possible.

 

2 or 3 ports in the rear of the chamber – these may be used for loading and unloading the PickUpLine Consider how plates will be loaded and removed from the chamber.

 

An extra 1 or two ports nearer the loading bay may be useful. Servicing if an engineer is required to service the PIXL some things can be done by reaching in from the main access port. In the event of a breakdown, it may be necessary to remove the PIXL from the chamber.

 

Access to the electrical cabinet drawer – required to service or fault find. This is a drawer that slides out to the left of the PIXL and it is almost the full width of the PIXL.

WHAT FORMAT SOURCE PLATES DOES PIXL TAKE?

PIXL accepts Petri dishes (90mm, 120mm) as well as rectangular plates, multiwell plates (at the densities 6, 12 ,24 ,48, 96 and 384) and 120mm square plates.

Using PIXL’s re-array mode custom plate formats can be loaded; we have supported synthetic biologists wishing to load MALDI-TOF slides for example. We love creative applications – get in touch with your novel idea.

WHAT FORMAT TARGET PLATES DOES PIXL TAKE?

As standard, PIXL pins onto agar or into liquid, on SBS-format rectangular plates, including 96 and 384 PCR and multiwell plates — both shallow and deep.

Absolutely! ROTOR+ and PIXL are designed so that anyone can operate them in a matter of minutes! No need for a dedicated technician, laborious calibration and maintenance, or hours of training; our simple, touchscreen software guides you through your chosen protocol with ease. How many colonies can I array on a plate?

ROTOR+ can array as many as 6,144 colonies on a single PlusPlate (actually there is a paper proving it can pin to 24,576!) ROTOR+ also supports arraying at 96, 384, 768, and 1,536 densities. Multiple low-density plates may be combined into fewer high-density plates by offsetting the pins in a square matrix.

HOW FAST IS ROTOR+?

ROTOR+ uses disposable plastic pin pads, which removes the need for lengthy washing and sterilisation cycles between pinning. The robotic arm always selects the shortest path between source and target and moves very rapidly (up to 3 metres per second). Oh, and did we mention that it can replicate the entire yeast genome in <25 seconds?!

Does ROTOR+ support liquid transfer?

The strength of ROTOR+ is to pin colonies on agar plates, but it often first needs to collect cells from a liquid suspension. ROTOR+ can transfer liquid to or from multiwell plates using adhesion to special long pin RePads, transferring approximately 200 nl of liquid on each pin

ARE THE ROBOTS STERILE?

PIXL uses specially developed Pinpoint™ picking technology for a step-change in reliability and sterility. PickupLine comes gas sterilised, prepackaged in a sealed bag and is thermally sterilised again, just before picking. Pinpoint uses the freshly-cut end of a sterile PickupLine™ to transfer microbial colonies.

ROTOR+ uses disposable plastic replica plating pads that come in sealed bags, which are then gamma irradiated, ready to be used.

Both robots have a built-in UV light that disinfects the working environment in between uses.
Together, the robots have a cross contamination rate of <0.04%.

CAN ROTOR+ PIXL COPE WITH UNEVEN AGAR?

Yes, totally! Agar is rarely flat. It’s probably the biggest challenge in colony picking automation and one of the areas where these robots really dominate. The gimbal-mount colony manipulating printhead on ROTOR+ senses the surface of the agar, applying just 1 gram of load, ensuring tolerance to different agar heights and surface variability without splashing cells everywhere or missing colonies (those of you who know, know!) The gimbal-mount also allows the print head to morph onto the agar surface even if it is at an angle.

By design, PIXL’s Pinpoint™ picking technology morphs to differences in agar height and carefully regulates the contact pressure. It happily maintains picking accuracy and repeatability across uneven agar surfaces and easily copes with agar height variations from plate to plate. Oh, and all of this is completely automated on every pick!

WHICH FLUORESCENT MARKERS CAN I SELECT FOR?

PIXL will quantify and allow selection for wtGFP, sfGFP, mCherry, tagBFP and Venus as standard, these proteins have been tested in house. Get in touch if you are interested in checking compatibility with a different marker. Because PIXL is a colony picker and not a microscope, it will not detect sub-cellular or very low levels of expression.
