DNA damage response pathways have been studied extensively in the budding yeast Saccharomyces cerevisiae, yet new genes with roles in theDNA damage response are still being identified. In this chapter we describe the use of functional genomic approaches in the identification of DNAdamage response genes and pathways. These techniques take advantage of the S. cerevisiae gene deletion mutant collection, either as an ordered array or as a pool, and can be automated for high throughput.
