High-Throughput Screening: ROTOR HDA

Genome-wide screening is easy when you have the right tools. You need a collection library – which we don’t provide (but we can tell you who does)! And you need a precision method of pinning colonies – this is why we created the ROTOR.

 


How does it work?

The ROTOR uses a robotic arm and sterile, ready-to-use replica plating pads to pick up samples of cells from culture plates. The robotic arm then moves to a new ‘target’ plate and deposits the cells. The operator selects and uses an appropriate program from a simple, drop-down menu.

 

What is it?

The ROTOR is capable of pinning nearly a million colonies per hour. Making it the fastest and most powerful colony manipulation robot in the world. It is essential for any large-scale microbial functional or chemical genomic studies.

 

What does it do?

This easy-to-use, compact bench-top robot is elegantly designed for the picking and re-plating of yeast, fungi, bacteria and algae colonies at densities 96, 384, 1536 and 6144. Using the 6144 RePads, the ROTOR can replicate the entire yeast deletion library in quadruplicate in less than 20 seconds! Reagent sets such as deletion mutant collections and the complete set of cloned yeast genes can be utilised for high-throughput 2-hybrid, synthetic genetic array, phenotypic and chemical-genetic analyses.

What can I use it for?

Duplicate and back up large libraries of yeast, fungi, bacteria and algae.

Re-plate cells from liquid to agar and vice versa.

FAQS

What plates can I use?

For precise replication at all densities, we recommend using Singer high quality PlusPlates with increased plating area. The ROTOR HDA will also accept multi-well, microtitre plates (96 and 384) and SBS standard trays.

Can I use Omni Trays and other SBS-format plates?

Yes. However, Omni Trays and other SBS-format plates will not be compatible with short pin RePads. This means that you will only be able to pin colonies at 96- or 384- densities using these plates. If you would like to screen at 1,536-density and beyond, you would need to use the Singer PlusPlates.

How does ROTOR combine multiple low-density plates into a high-density plate?

The ROTOR combines four low-density plates into one high-density plate by offsetting the pins in a square matrix. See pinning diagrams here.

Is the ROTOR easy to use?

Absolutely! The ROTOR HDA is designed so anyone can operate the robot in a matter of minutes! No need for a dedicated technician or hours of training; our simple, touchscreen software guides you through your chosen protocol with ease.

Why is the ROTOR so fast?

The ROTOR HDA uses disposable plastic pin pads, which removes the need for lengthy washing and sterilization cycles between pinning.
The robotic arm moves very rapidly (up to 3 metres per second) and always selects the shortest path between source and target.
On removing the pinned plates you can immediately see the impressions the pin heads make and know for sure that you can expect the experiment to be successful if all other factors are right. If a slight modification needs to be made it can be done so using the simple controls such as pin pressure for an unevenly poured or dried out plate. It’s press and play not press and prey.

How many colonies can I array on a plate?

The ROTOR can array as many as 6,144 colonies on a single PlusPlate! ROTOR also supports arraying at 96, 384, 768, and 1,536 densities. Multiple low-density plates may be combined into fewer high-density plates.

Does the ROTOR support liquid transfer?

Although the strength of the ROTOR is to pin colonies on agar plates, it does offer some liquid transfer capabilities. By taking advantage of the capillary action on the surface of the pins, the ROTOR can transfer liquid using long pin RePads and transfer approximately 200 nl of liquid on each pin.
